Peptide information
Retatrutide is a 39 amino acid single peptide with triple agonist activity at the glucagon receptor (GCGR), glucose-dependent insulinotropic polypeptide receptor (GIPR), and glucagon-like peptide-1 receptor (GLP-1R). The peptide backbone contains three non-coded amino acid residues at positions 2, 20, and 13. Stability is increased by the amino acid sequence Aib2, or amino isobutyric acid, which is an inhibitor of DPP4 cleavage. A distinct amino acid sequence called Aib20 affects the pharmacokinetic profile, optimal GIP activity, and developability. The third amino acid sequence necessary for normal GIP activity and glucagon levels is αMeL13, or α-methyl-L-leucine. The backbone is conjugated to a C20 fatty diacid moiety at position 17. Retatrutide was discovered by Eli Lilly and Company and is currently in Phase 3 clinical trials for obesity and type 2 diabetes research.
